ANALISIS BENTUK MORFOLOGI SEL DARAH MERAH MENGGUNAKAN PEWARNA GIEMSA DENGAN VARIASI WAKTU FIKSASI MENGGUNAKAN METANOL

Objective: To analyze the effect of Methanol fixation time variation on erythrocyte morphology and determine the optimal fixation duration.Methods: A laboratory experimental study was conducted on 30 blood samples from students, with Methanol fixation times of 2, 5, 10, and 15 minutes. Giemsa staining was applied, followed by microscopic observation. Data were analyzed using the Chi-Square test.Results: There was a significant difference in erythrocyte morphology based on fixation time variation (p = 0.016). A 2-minute fixation resulted in the most optimal morphology, while the 15-minute group showed increased abnormal cell shapes.
